Abstract
Ocotea quixos (Lam.) Kosterm. (Ishpingo) is a tree that grows in humid tropical forests of South America (310–1200 m above sea level) and is used medicinally and as a condiment, as it possesses essential oils with antiplatelet and antithrombotic properties, among others. In Ecuador, wild populations of Ishpingo are declining due to ecosystem transformation, deforestation, and clonal cultivation. This study compared the genetic variability of three chloroplast DNA regions (matK, rbcLa, and psbA trnH) and one nuclear ITS region from Ishpingo obtained from five provinces of the Ecuadorian Amazon. Based on the alignments obtained, phylogeny was performed using the maximum likelihood criterion, with a bootstrap of 1000 replicates and Jukes-Cantor. Genetic variability was determined using the DNA conservation coefficient and Nei's nucleotide variability coefficient (π). The nucleotide variability of the matK, rbcLa, and psbA-trnH regions was low (π ≤ 0.005) due to their high degree of conservation. The ITS region showed greater variability (π = 0.11), grouping the analyzed individuals into two clades. The results for the ITS region may be affected by the homoplasy present in this region. The analyzed regions were not useful for studying genetic variability in Ishpingo. Therefore, it is suggested that new regions of chloroplast and nuclear DNA be analyzed to allow for population genetic studies of this species in Ecuador.
