La presencia de la UPS en la ciudad de Quito inicia en 1994 con la creación de la Facultad de Ciencias Humanas y Sociales conformada por tres programas de pregrado: Psicopedagogía, Antropología Aplicada y Teología Pastoral. Sin embargo, es en 1995 cuando se crea y oficializa la Sede Quito. La Sede nace con las siguientes facultades: Ciencias de la Educación, Ciencias Humanas y Sociales, Ciencias Religiosas, Ciencias del Desarrollo y Ciencias Técnicas. Orientados a atender las demandas de formación de la población quiteña pero también de otras localidades del país: Cayambe, Zumbahua, Ambato, Riobamba, Santo Domingo, Esmeraldas, Puyo, Macas, entre otros, por medio de programas semipresenciales y a distancia.
